    宽体沙鳅与中华沙鳅同工酶比较研究

    Comparation of the Four Isozymes in Sinibotia reevesae and Sinibotia superciliaris

    • 摘要: 通过聚丙烯酰胺凝胶电泳法对宽体沙鳅 Sinibotia reevesae和中华沙鳅 Sinibotia superciliaris成体的心脏、肝胰脏、肠、肾、肌肉、脑、眼7种组织中的酯酶( EST)、过氧化氢酶(CAT)、乳酸脱氢酶( LDH)、苹果酸脱氢酶(MDH)共4种同工酶的活性和类型进行了比较研究.结果表明: (1)在宽体沙鳅和中华沙鳅7种组织中检测到6种酯酶同工酶, 2种过氧化氢酶同工酶, 4种乳酸脱氢酶同工酶, 8种苹果酸脱氢酶同工酶表达; (2)在7种组织中,各同工酶表现出明显组织特异性,这与同工酶的功能密切相关; ( 3)在两种中华沙鳅属鱼类中, 4种同工酶酶带的分布具有一定相似性和差异性,体现在宽体沙鳅和中华沙鳅 EST-1、 EST-2 同工酶表达相似性较高, CAT同工酶在肝胰脏和中肾组织表达较强, LDH 同工酶酶谱相对简单,线粒体型 MDH 同工酶在两种鱼类的各组织中表达差异较大.

       

      Abstract: Four isozymes(EST, CAT, LDH and MDH) in seven tissues(gill, heart, hepatopancreas, intestines,
      kidney, muscle, brain and eye) of Sinibotia reevesaeand S. superciliaris were comparatively studied using the method of poly- acrylamide gel electrophoresis. The results showed that, 1. It was found that there exist six esterase isozyme bands, two cata- lase isozyme bands, four lactate dehydrogenase isozyme bands, eight malate dehydrogenase isozyme bands; 2. Each isozyme displays an apparent unique tissue-specific property in seven tissues of the two fishes and is closely related to its physiological function; 3. There were similarities and differences between the four isozyme bands of two Sinibotia genus of fishes. The grea- ter similarity was found in EST-1 and EST-2 isozyme expression between S. reevesae and S. superciliaris. The expression of CAT isozyme in hepatopancreas and kidney of two Sinibotia genus fishes was stronger than in other tissues. The expression of LDH isozyme was relatively simple in terms of zymogram. The expression of mitochondrial MDH isozyme was found to dis- play a rather huge difference in the same tissues of S. reevesaeand S. superciliaris.
