基于SSR的云南省魔芋属两个野生种的遗传多样性评价

Genetic diversity assessment of two wild species inAmorphophallusgenus from Yunnan Province

  • 摘要:魔芋属( Amorphophallus)是中国的特色经济作物,当前对魔芋种质资源的遗传评价不足,影响了野生资源的利用.利用SSR分子标记技术对云南省自然分布的滇魔芋( A. yunnanensis)和东京魔芋( A. tonkinensis)的两个野生种群的遗传多样性进行了分析.结果表明,选用的SSR位点的多态性较好,有效等位基因数(Ne)为1.240~3.816.滇魔芋和东京魔芋的观察杂合度(Ho)分别为0.581和0.369,期望杂合度(He)分别为0.526和0.349.所有遗传多样性参数都表明滇魔芋的遗传多样性高于东京魔芋,这可能与滇魔芋的分布区域较广有关.两个野生种之间存在中等水平的遗传分化(Fst = 0.132),群体遗传结构分析可以将两个种群完全区分出来.但两个魔芋种间存在一定程度的杂交,说明二者亲缘关系较近.研究结果对制定魔芋属的种间杂交策略及野生资源保护措施具有一定参考价值.

    Abstract:Species in the Amorphophallusgenus have long been cultivated as cash crops in China. However, the lack of genetic assessment of wild species has hindered the utilization of wild germplasm resources. In this study, the genetic diversity of two wild species of Amorphophallusgenus distributed in Yunnan province ( A. yunnanensisand A. tonkinensis) was analyzed using simple sequence repeat (SSR) markers. The results showed that the polymorphisms of SSR loci were high, with the effective allele number ranging from 1.240 to 3.816. The observed heterozygosity (Ho) and expected heterozygosity (He) were 0.581 and 0.526 for A. yunnanensis, as well as 0.369 and 0.349 for A. tonkinensis, respectively. All genetic diversity indices suggested that the genetic diversity of A. yunnanensiswas higher than that of A. tonkinensis, which might be associated with the wider distribution range of A. yunnanensis. A moderate level of genetic differentiation between the two species was revealed (Fst = 0.132), and the genetic structure analyses could completely separate these two species from each other. Nevertheless, hybridizations between A. yunnanensisand A. tonkinensis were also detected, which indicated a relatively close relationship between the two species. The findings of our study could help the interspecific hybridization and the protection of wild resources of Amorphophallus.
